Monoclonal antibody titer assays are a growing bottleneck in cell culture process development. The current “gold standard” method – protein A affinity HPLC – is reliable, selective, sensitive and precise, but throughput is limited. Other methods are either complex to perform or require dedicated instrumentation. The AssayMAP® MAb Titer assay was developed to utilize the same method and chemistry as affinity HPLC, while delivering dramatically improved throughput by means of parallel sample processing, without special instruments or equipment.

What’s the advantage? 100-fold increase in throughput achieved using standard laboratory equipment and your current validated reagents. All you need is the AssayMAP MAb Titer kit, a microplate centrifuge, your current HPLC buffers, a micropipettor, plate reader and the basic protocol. The same assay also can be fully automated on conventional robotic liquid handlers.

• Utilizes the same operating principle and chemistry as the “gold standard” protein A affinity HPLC.
• Dynamic range of <10 to >10,000 μg IgG/mL, without sample dilution.
• Runs effectively with small numbers of samples, but can have throughput of >384 samples per hour.
• Can be run manually using only a micropipettor, microplate centrifuge and plate reader or fully automated using widely available equipment.

The heart of the assay is the cartridge containing a 5 µL bed packed with a rapid mass transport protein A affinity resin. The highly versatile cartridges are run in parallel as ultra-micro spin columns. Controlled flow provided by specially engineered labware enables precise, quantitative binding and elution. The cartridges, when handled properly, are reusable 5 to 10 times, making them simple to use, efficient and cost effective. A special cartridge rack in SBS compatible microplate format enables parallel processing of 1 – 96 samples per plate in any combination.

AssayMAP MAb Titer Kit Standard Curves

A 2x serial dilution of hIgG was made from 5000 µg/mL downward, and 10, 30 and 100 µL samples of each standard were run. Readout was done using both OD 280 nm and with Coomassie blue reagent. The OD 280 nm is highly linear up to just over 100 µg hIgG, then flattens out. This is due to saturation of the cartridge binding capacity. The Coomassie blue readout is ~10X more sensitive, but becomes non-linear above ~15 µg hIgG, due to saturation of the dye binding. If the full range of sample volumes and the two readout methods are utilized, the assay has an effective range of 10 to 10,000 µg/mL.

Getting Started ► We offer a starter kit for first time users. This kit includes 24 reusable cartridges, a comprehensive protocol and two additional empty racks and receiver plates for managing centrifuge balance and simplifying pipetting.

Qualification ► After you have an initial start-up kit, all you need for scale-up is more cartridges offered in convenient packs that contain 96 qty cartridge-only racks. No duplicate protocols, no extra racks.

Implementation ► At some point you will want to build a solution that fits your specific needs. We offer bulk purchase options to personalize your order and provide additional savings.
