A High Throughput, High Precision Bind and Elute Assay

The AssayMAP MAb Titer assay offers a means for measuring antibody titer in bioprocess samples. Using the same Protein A chemistry employed in conventional affinity HPLC assays, the AssayMAP Cartridges offer parallel sample processing in just minutes. What’s the advantage? 100-fold increase in throughput achieved using standard laboratory equipment and your current validated reagents. All you need is this starter kit, a microplate centrifuge, your current HPLC buffers, a micropipettor, plate reader and the basic five-step protocol. The same assay also can be fully automated on conventional robotic liquid handlers.

The patent-pending cartridge technology allows you to perform the assay without the complexity and limited throughput of HPLC. The optimized 50 µL bed geometry and rapid mass transport beads enables quantitative results to be obtained in a spin column format miniaturized to work with microplates. A special cartridge rack in SBS compatible microplate format enables parallel processing of 1 – 96 samples per plate in any combination.

Principally, it is the same as your current HPLC – but with far greater throughput.

Monoclonal antibody titer assays are a known bottleneck in process development. The AssayMAP technology offers a very low risk solution to break through that bottleneck. AssayMAP MAb Titer Kit Standard Curves

The MAb Titer Assay directly quantifies IgG product concentration from less than 10 to greater than 5000 µg/mL, without requiring sample dilution. Sample volume can be varied from 5 to 200 µL. Higher IgG product concentrations are quantified directly by A₂₈₀ measurement (A) and lower concentrations are quantified by addition of a colorimetric protein reagent such as Coomassie Blue in a Bradford Assay (B).

Getting Started: We offer a starter kit for first time users. This kit includes one rack of cartridges (96 qty), a receiver plate, a comprehensive protocol and two additional empty racks and receiver plates for managing centrifuge balance and simplifying pipetting.

Scaling up: After you have an initial start-up kit, all you need for scale-up is more cartridges offered in convenient packs that contain 96 qty cartridge-only racks. No duplicate protocols, no extra racks.
