N-glycan profiles of MAbs can affect immunogenicity, pharmacokinetics and efficacy. Profiles are profoundly affected by cell culture conditions, but current manual sample preparation methods require days to complete. The AssayMAP cartridge selectively binds MAb from a crude cell culture sample. After impurities are removed, bound MAb is reacted with enzyme to selectively cleave N-glycan groups, which are fluorescently labeled and cleaned up with a separate hydrophilic interaction cartridge. This complex workflow can be easily automated and completed in 3 hours or less.
