Flexible Workflows
AssayMAP cartridges can be used as modular building blocks, assembled in a wide variety of ways to create both simple and complex assay workflows for a wide range of applications. The following examples show how a single cartridge type can be used in variety of different ways. In this case a selective antibody-binding cartridge (such as protein A) is used in three different critical assays in upstream process development for MAbs.

| Cell line selection requires highly sensitive quantitation of the product MAb using low sample volumes. ELISAs are most often used, but are imprecise, complex and time-consuming. The AssayMAP cartridge selectively binds the product MAb and a fluorescently-labeled antibody-binding reagent and then forms a “sandwich”. The MAb and labeled reagent are eluted and quantified in a fluorescence plate reader. | |

| Scaleup optimization of the cell culture process requires far less sensitivity, but high precision is a must. The “gold standard” method is affinity HPLC, but the serial nature of this method has made throughput a major challenge. The AssayMAP cartridge again selectively binds the product MAb, which (after washing away impurities) is selectively eluted and accurately quantified using a UV plate reader. Precision is comparable to HPLC with 100 times the throughput, even using manual liquid handling. | |

| N-glycan profiles of MAbs can affect immunogenicity, pharmacokinetics and efficacy. Profiles are profoundly affected by cell culture conditions, but current manual sample preparation methods require days to complete. The AssayMAP cartridge selectively binds MAb from a crude cell culture sample. After impurities are removed, bound MAb is reacted with enzyme to selectively cleave N-glycan groups, which are fluorescently labeled and cleaned up with a separate hydrophilic interaction cartridge. This complex workflow can be easily automated and completed in 3 hours or less. | |
